Using V 2.3.0.0
When the label properties Interline = 1 and Font size = 3 and using "Test" PDF the call sign font size is a bit to large and bleeds into the QSO detail header line. In addition the length of the sample call sign is to long for the font size so it wraps around into two lines.

Attached are the settings I have for my label Page Layout and Label Properties. Note my margin settings are set to work for USA-sized paper and the Avery 8160 labels. There is another Avery label with a different ID number, but is the same size. One is for laser printers and one is for ink-jet printers.

You did not note that in your original post so I had no idea what you were printing to. Ergo, I provided what I've used at my station.
Anyway ... no matter ... as you just have to trial & error settings to see how best to get them situated for your specific printing device.
I do many sample tests to regular paper and then overlay that on the template I'm wanting to use. May be somewhat similar with using the Dymo before you have to print a "live" label.
Best wishes with your tests and once you get your template configured save it as Jurgen has noted so it doesn't get over-written with V2 updates.
